Versatility and dynamics of transcriptional responses to Notch

Abstract
Transcriptional changes elicited by signalling are key to many developmental and pathological processes. Often these are considered only in terms of simple static patterns of on/off gene expression, I will discuss two ongoing projects that highlight the importance of looking beyond. First, investigating the maintenance of Drosophila muscle progenitors we discovered that signaling switches the type of RNA produced; Notch activity directs muscle progenitors to produce an alternate zfh1/ZEB1 RNA -isoform that protects them from differentiation. Second, using the MS2 /MCP system to live-image transcription in real-time from two Notch responsive enhancers we reveal that signaling elicits highly dynamic profiles of transcription and how these differ according to other spatial inputs.
