The evaluation of lyssavirus cross-neutralising antibody responses using lentiviral pseudotypes.

Abstract
We have analysed cross-neutralisation between rabies virus, CVS -11, and two European bat lyssaviruses, EBLV -1 and -2, using lentiviral pseudotypes as antigen vectors. This development of a robust microassay that can be performed outside of high containment facilities will aid on-going surveillance and vaccine studies. Tests on blinded sera from rabies virus vaccine recipients revealed that the CVS -11 pseudotype assay had 100% concordance with the current “gold standard” fluorescent antibody virus neutralisation test, and neutralising titres strongly correlated. Pseudotypes incorporating the G-protein from different lyssavirus genotypes were used to analyse antibody titres with respect to G-protein identity. The results revealed that the relative neutralisation titres correlated broadly with the degree of G-protein diversity.
