Abstract

A challenge in bioelectronic medicine is to restore neurological functions lost to disease and in particular to develop models of synthetic microcircuits that accurately replicate the adaptation of neurons and central pattern generators to physiological feedback.

This talk shall cover the data assimilation methods we have developed to transfer information from in-vitro electrophysiological recordings of biological neurons (brain and CNS ) to neuron-based conductance models. Completed models are validated by twin experiments and by predicting the membrane voltage oscillations induced by a wide range of current simulation protocols. I shall discuss various aspects of the parameter inference problem in particular the search for unique parameter solutions and criteria for constraining those in the context of ill-posed problems. I shall report on the optimization of VLSI models of respiratory neurons and their implementation in implantable analog silicon microcircuits. These microcircuits have been successfully used to restore cardio-respiratory coupling in heart failure and to provide a therapy in two animal models of disease.
