DESCRIPTION:*ABSTRACT*:\nA cancer genome contains the historic mutagenic a
 ctivity that has occurred throughout the development of a tumour. While dr
 iver mutations were the main focus of cancer research for a long time\, pa
 ssenger mutational signatures - the imprints of DNA damage and DNA repair 
 processes that have been operative during tumorigenesis - are also biologi
 cally informative. In this lecture\, I provide a synopsis of this concept\
 , describe the insights that we have gained through computational analysis
  of human cancer data and through experiments in cell-based systems\, and 
 showcase how we have developed the concept into clinical applications. I w
 ill present unpublished data on individual whole-genome-sequenced cases as
  well as a population-wide study to demonstrate how we aim to translate th
 ese ideas towards clinical utility\, as early as possible in a patient’s
  diagnostic journey.
