DESCRIPTION:Mouse Hox gene enhancer analysis has previously been conducted
  in embryos transgenic for Hox/lacZ reporter constructs. Here I show how a
 t least some of these studies can be carried out in cultured HepG2 cells u
 sing Hox/luciferase reporters and a doxycycline-inducible expression syste
 m. In particular\, I show the following: 1) Hoxd11 enhancer is activated b
 y Gdf11/Smad signalling\, 2) Hoxb4\, b8 and a7 enhancers are activated by 
 Cdx1 protein\, and 3) Hoxc8 enhancer is activated by the synergistic actio
 n of Gdf11/Smad signalling and Cdx proteins. Hox enhancer analysis in cell
  culture reduces mouse usage\, requires low cost and skill\, permits rapid
  assessment of putative enhancer binding motifs\, and provides abundant ma
 terial for DNA/transcription factor binding studies.
