DESCRIPTION:Damage in brain blood vessels contributes to cognitive impairm
ent in most cases of Alzheimer’s disease. Vascular dementia is the secon
d most common dementia subtype after Alzheimer’s. Unlike Alzheimer’s d
isease where culprit molecules are well established\, notably APP-derived
beta-amyloid and hyper-phosphorylated tau protein\, brain vascular disease
lacks molecular targets and urgently needs better chemistry.\n\nAtticus H
ainsworth’s group test molecular hypotheses in human brain tissue and in
experimental models. We are especially interested in brain endothelial ce
lls\, blood-brain barrier dysfunction and white matter lesions. We recentl
y completed a phase II clinical trial\, testing the phosphodiesterase inhi
bitor drug tadalafil for possible repurposing in vascular dementia.
