DESCRIPTION:Building better models of cellular pathways is one of the majo
r\nchallenges of systems biology and functional genomics. There is a\nneed
for methods to build on established expert knowledge and\nreconcile it wi
th results of high-throughput studies. Available data\nsources are very he
terogeneous and need to be combined in a way\nspecific for the part of the
pathway they are most informative for.\nAlso\, some data sources like pro
tein domains or phosphorylation\nstates have so far been under-utilized fo
 r inferring pathway\nstructure. Here we present a compartment-specific str
ategy to refine a given\npathway hypothesis from a compendium of heterogen
eous data sources\,\nwhich includes novel methodology to integrate protein
attributes. We\ndemonstrate the power of our approach in a case study in
S.\ncerevisiae and show that it can assist experimentalists in planning\nf
uture studies by proposing testable extensions and refinements of\na give
 n pathway hypotheses.
