DESCRIPTION:Proteins have multiple conformations with distinct functional 
 roles. How the conformations interconvert to regulate function is an activ
 e area of research. We have been studying two classes of DNA unwinding enz
 ymes\, CRISPR Cas9 and helicases\, and how their activities are regulated 
 by conformational control. We have used the mechanistic knowledge to engin
 eer new variants of the enzymes that can be activated through light or che
 mical crosslinking. I will describe several biotechnological applications\
 , including precision genome manipulation in time and space\, co-transcrip
 tional RNA folding\, isothermal DNA amplification and genome imaging.
