DESCRIPTION:In the time it takes you to read this sentence\, your body wil
l produce millions of new cells. It is critical that each of them copied t
heir genome accurately\; errors in DNA replication are a hallmark of cance
r and other genetic diseases. My group is developing artificial intelligen
ce software that measures the movement of DNA replication forks from Oxfor
d Nanopore sequencing data. Using AI in this way has allowed us to detect
sites of DNA replication initiation and fork stalling with a spatial resol
ution and throughput that exceeds previous microscopy-based methods by sev
eral orders of magnitude. I will discuss our progress using this technolog
y in human cancer cells and parasites\, where we are working on how to bet
ter utilise faulty DNA replication as a therapeutic target. I will also di
scuss process algebra-based modelling and simulation techniques we are dev
eloping to help guide our experiments.
