DESCRIPTION:Loop extrusion has emerged as a prominent hypothesis for how r
 ing-shaped SMC complexes shape chromosomes and participate in genome trans
 actions. Single molecule in vitro observations have yielded stunning movie
 s of this process. When not extruding loops\, SMC complexes are known to t
 opologically capture one or more DNAs\, offering an alternative explanatio
 n for how they might shape the genome. Here we propose a molecular framewo
 rk for how topological DNA entry into the cohesin ring involves a DNA moti
 on that\, under certain conditions\, repeats itself to elicit loop extrusi
 on. We then explore whether topological loop capture\, or loop extrusion\,
  is more likely to explain physiological SMC complex functions. As an exam
 ple\, we will study how mitotic chromosomes gain shape during cell divisio
 ns.
