DESCRIPTION:R-loops are three-stranded nucleic acid structures that form c
o-transcriptionally and which are comprised of an RNA-DNA hybrid and a dis
placed strand of ssDNA. Although R-loops have important functions in cells
\, their dysregulation can also lead to genome instability. We recently sh
owed that a surprising consequence of dysregulated R-loop metabolism is th
e release of RNA-DNA hybrid fragments into the cell and subsequent sensing
of these hybrids by innate immune receptors. In this seminar I will descr
ibe our recent studies demonstrating how these pathological R-loops arise
and features that cause them to threaten genome stability.
