DESCRIPTION:Abstract:  \n\nMorphogenesis relies on the precise coordinat
 ion of single-cell behaviors to build complex tissues. Intestinal developm
 ent exemplifies this process\, as crypt formation emerges from tightly reg
 ulated biochemical and mechanical cues. Using organoid models combined wit
 h in vivo studies\, we dissect how crypt morphogenesis is initiated by act
 omyosin-driven apical constriction and accelerated by osmotic forces. We i
 dentify a critical mechanochemical feedback loop mediated by calcium-depen
 dent cytosolic phospholipase A2 (cPLA2)\, which senses mechano-osmotic cha
 nges and triggers robust crypt formation via arachidonic acid production a
 nd myosin relocalization. Together\, our work reveals how cells integrate 
 mechanical and osmotic signals to orchestrate irreversible tissue-scale tr
 ansformations during development.
