DESCRIPTION:A major component of the response of cells to changing conditi
 ons is a\nshift of the transcriptome to a new state which may be more adeq
 uate for\nfacing the new environment. The time dependence of this shift is
  of\nconsiderable interest and is often inferred from measurements of mRNA
 \nconcentrations. \n\nWe formulate a refined model for the dynamics of pre
 -mRNA and mRNA\nconcentrations and show that the temporal profiles of pre-
 mRNA are much\nmore suitable than those of mRNA for reconstruction of the 
 transcript\nproduction profiles. By monitoring time-dependent changes of p
 re-mRNA\nabundance in MCF10A cells stimulated by EGF\, we show that in gen
 eral\,\nproduction profiles exhibit different kinds of interesting and\nun
 expected forms. In order to increase expression of specific genes to\nsome
  high desired level in a short time\, cells use a strategy of\nproduction 
 overshoot for a relatively brief interval. \n\nA genome wide view shows wh
 ile these production profiles are closely\nreflected by the time dependenc
 e of the pre-mRNA\, the corresponding mRNA\nprofiles (which also exhibit u
 nexpected complexity) are often\nuncorrelated with transcript production.
