DESCRIPTION:My laboratory has made significant advancements in understandi
 ng the unconventional roles of uncharacterized and understudied protein ki
 nases and pseudokinases. Over the past ten years\, we have uncovered unexp
 ected biochemical functions within this protein family\, including AMPylat
 ion\, polyglutamylation\, and mRNA capping via RNAylation. Our approach em
 ploys a multifaceted strategy\, beginning with the bioinformatic identific
 ation of atypical family members. We then use biochemistry\, structural bi
 ology\, and mass spectrometry to reveal unique functions and previously un
 discovered enzymatic activities that could not have been predicted a prior
 i.\n\nWhile traditional sequence-based bioinformatics approaches have been
  highly effective in identifying new kinase families\, integrating AI-driv
 en tools like AlphaFold into our experimental pipeline has been a transfor
 mative advancement. In this lecture\, I will present two recent discoverie
 s from my laboratory that harnessed AI to identify orphan kinases in the e
 ther lipid and isoprenoid salvage pathways.
