DESCRIPTION:Electron tomography of frozen-hydrated specimens\, without che
 mical fixation\, staining\, or dehydration\, allows us to study the 3-D st
 ructure of cells and tissue in a near-native state.  Preparation and 3-D i
 maging of such specimens is challenging.  Developments in the application 
 of cryo-ultramicrotomy and cryo-FIB\, as well as TEM imaging using zero-lo
 ss energy filtration and Zernike phase contrast\, will be discussed.
