DESCRIPTION:Most cellular responses to extracellular stimuli have a common
  component of regulation arising from selective\nrecruitment of a network 
 of signalling complexes to membranes. Â However\, studying these systems
  remains a daunting\ntask. Â We have made unprecedented progress in unde
 rstanding these systems by applying a synthesis of hydrogen deuterium\nexc
 hange mass spectrometry (HDX-MS)\, X-ray crystallography and FRET spectros
 copy. Our application of this synthesis to\nphosphoinositide 3-kinase (PI3
 K) signalling represents one of the largest (> 200 kDa) complexes studied 
 using DXMS\, and\nelucidates novel aspects of PI3K regulation downstream o
 f GPCRs as well as a unifying mechanism of how diverse\noncogenic PI3K mut
 ants upregulate activity.
