DESCRIPTION:In vitro\, 3D cultures are limited by oxygen and nutrient diff
usion rates resulting in necrosis at the centre of large scaffolds. Mass
transport in these cultures can be controlled to a large extent by use of
dynamic bioreactors. The bioreactor configuration and operation is import
ant in the success of the culture. Previous work has showed biaxial rotat
ion was more successful for in vitro tissue generation than single axis bu
t did not describe the mechanism underlying this effect.\n\nIn this presen
tation\, a description of the current state of the bioreactor field for bo
ne growth is described before the key design requirements of a new low cos
t design are identified. Finally to investigate the parameters that are c
ritical when designing a rotating bioreactor\, two low-cost\, adaptable an
d open source bioreactor systems are presented and control systems verifie
d for rotational and positional control.
