DESCRIPTION:The regulatory cell state of founder stem cells during develop
 ment\, and those in the adult is determined by critical transcription fact
 ors. We and others have demonstrated that distinct gene regulatory network
 s operate in different anatomical locations to give rise to skeletal muscl
 e progenitor cells. Despite this diversity\, the core cell fate determinan
 ts\, Myf5\, Myod and Mrf4 regulate cell identity in all locations. Upstrea
 m of this progenitor population\, Notch signaling maintains the stem cell 
 population whilst allowing temporal cell identities to emerge. Cell hetero
 geneity in the adult muscle stem cell population is observed among quiesce
 nt and proliferating cells. We have isolated and characterised subpopulati
 ons of muscle stem cells during homeostasis and regeneration. Single cell 
 analysis provides insights into the pallette of behaviours of muscle stem 
 cells.
