DESCRIPTION:In my seminar I will discuss a series of studies from the lab
aimed at understanding the relationship between the structure of receptor-
ligand complexes and signaling. The main focus will be on our efforts to e
ngineer protein ligands\, using combinatorial biology\, to engage receptor
s in alternative fashions in order to alter cell subset specificity and/or
signaling amplitude. We have found that signaling by dimeric cell surface
receptors\, principally cytokine receptors\, is surprisingly 'tunable' th
rough ligand engineering. I will also discuss some recent attempts to app
ly these principles in a GPCR system.
