DESCRIPTION:In recent years\, we have combined DNA nanotechnology with sin
 gle-molecule\nfluorescence to create functional single-molecule devices su
 ch as nanoscopic\nrulers for superresolution microscopy and energy transfe
 r switches1-4. DNA\norigamis are also used to improve single-molecule dete
 ction by fluorescence\nenhancement with nanoantennas or by single-molecule
  placement in zeromode\nwaveguides using nanoadapters5\, 6. Especially\, f
 luorescence enhancement with\nself-assembled nanoparticles holds great pot
 ential for single-molecule\ndetection at higher concentrations but also fo
 r diagnostic applications7. We\nwill discuss recent advancement in fluores
 cence enhancement and how to\ndisentangle the complex factors that influen
 ce the fluorescence of single\nmolecules near metallic nanostructures8\, 9
 .
