DESCRIPTION:In recent years\, we have combined DNA nanotechnology with sin
 gle-molecule\nfluorescence to create functional single-molecule devices su
 ch as nanoscopic\nrulers for superresolution microscopy and energy transfe
 r switches[1-4]. DNA\norigamis are also used to improve single-molecule de
 tection by fluorescence\nenhancement with nanoantennas or by single-molecu
 le placement in zeromode\nwaveguides using nanoadapters[5\, 6]. Especially
 \, fluorescence enhancement with\nself-assembled nanoparticles holds great
  potential for single-molecule\ndetection at higher concentrations but als
 o for diagnostic applications [7]. We\nwill discuss recent advancement in 
 fluorescence enhancement and how to\ndisentangle the complex factors that 
 influence the fluorescence of single\nmolecules near metallic nanostructur
