DESCRIPTION:The questions of how genes are regulated remains fundamental e
 ven after many decades of intense study. Rather than just studying the lin
 ear\, one-dimensional sequence of DNA to inform us about regulatory mechan
 isms\, we can now investigate the complex 3-dimensional organisation of wh
 ole genomes. It has become clear that this organisation is non-random and 
 highly dynamic. To address new questions in genome architecture\, we are t
 aking a systems biology approach\, combining the bioinformatic determinati
 on of chromatin states with quantitative experiments and dynamic\, stochas
 tic models of whole genome organisation.  Comparing these results with our
  experimental data\, this has led us to understand how biophysical propert
 ies of the chromatin fibre lead to significant and biologically relevant s
 elf-organisation of the genome.
