DESCRIPTION:To systematically create complex yet reliable circuits\, elec
trical engineers use digital logic\, wherein gates and subcircuits are co
mposed modularly and signal restoration prevents signal degradation. We ha
ve adapted such engineering ideas to the design and experimental implement
ation of DNA-based digital logic circuits that operate in an aqueous envir
onment. We demonstrate AND\, OR\, and NOT gates\, signal restoration\, amp
lification\, feedback\, and composition of logic gates into multilayered c
ircuits. Biological nucleic acids such as microRNAs can serve as inputs\,
and circuits work reliably in a background of unrelated nucleic acids\, su
ggesting applications to the analysis of complex biological samples.
