Super-resolution microscopy describes a set of relatively recent microscopy techniques which allows to circumvent the optical resolution limit of traditional fluorescence imaging. Various implementations are available like STED , PALM/STORM, SIM , Photon reassignment, using different principles to achieve this. Other imaging parameters like imaging depth, temporal resolution or multi-color imaging are affected to varying degrees depending on the particular technique. During this presentation I will give an overview over current state-of-the-art super-resolution microscopy techniques available for the expert and non-expert researchers for performing high-resolution microscopy, with an emphasis on STED microscopy. I will discuss advantages of a number of methods and highlight applications which can benefit from super-resolution techniques. Practical considerations for using the available STED microscope in the department of Genetics will also be included.
