Conformational control: From single molecules to biotechnologies

Abstract
Proteins have multiple conformations with distinct functional roles. How the conformations interconvert to regulate function is an active area of research. We have been studying two classes of DNA unwinding enzymes, CRISPR Cas9 and helicases, and how their activities are regulated by conformational control. We have used the mechanistic knowledge to engineer new variants of the enzymes that can be activated through light or chemical crosslinking. I will describe several biotechnological applications, including precision genome manipulation in time and space, co-transcriptional RNA folding, isothermal DNA amplification and genome imaging.
