The awakening of the genome –modelling embryonic genome activation programs in vitro

Abstract
The activation of the zygotic genome (ZGA) marks the first major wave of transcription in the developing embryo and is crucial for developmental progression. To identify the molecular details of this process, we use stem cells to model ZGA in vitro. Using novel single cell sequencing technologies in naïve embryonic stem cells, we identified human 8-cell like cells (8CLCs), that resemble the human 8-cell embryo at the time of zygotic genome activation. 8CLCs represent a unique novel model system to study processes regulating human ZGA in vitro and will provide critical insights into early events in human embryogenesis in vivo.
