Chromosome Architecture: Lifting the Lid on Loop Extrusion

Abstract
Loop extrusion has emerged as a prominent hypothesis for how ring-shaped SMC complexes shape chromosomes and participate in genome transactions. Single molecule in vitro observations have yielded stunning movies of this process. When not extruding loops, SMC complexes are known to topologically capture one or more DNAs, offering an alternative explanation for how they might shape the genome. Here we propose a molecular framework for how topological DNA entry into the cohesin ring involves a DNA motion that, under certain conditions, repeats itself to elicit loop extrusion. We then explore whether topological loop capture, or loop extrusion, is more likely to explain physiological SMC complex functions. As an example, we will study how mitotic chromosomes gain shape during cell divisions.
