Abstract
Synapse loss is the strongest pathological correlate of cognitive decline in Alzheimer’s disease (AD) and genetic data strongly implicate both amyloid beta and changes in microglia in disease pathogenesis. In this talk, Prof Spires-Jones will talk about her work showing that (1) Amyloid beta (Ab) accumulates in synapses and contributes to synapse loss, (2) APOE4 and CLU risk genes associated with synapse loss, (3) Astrocytes and microglia ingest synapses in human AD brain, and (4) Blocking MFGE8 binding integrin receptors reduces human synapse phagocytosis by microglia and astrocytes.
