Pharmacological inhibition of protein aggregation within liquid condensates

Abstract
Protein misfolding and aggregation are characteristic of a wide range of neurodegenerative diseases. Emerging evidence suggests that protein aggregation may occur within liquid condensates formed through phase separation. This mechanism of aggregation creates new challenges and opportunities for drug discovery for protein misfolding diseases, which are otherwise still largely incurable. I will discuss the possibility of inhibiting the process of condensation-driven aggregation using small molecules. By using a chemical kinetics approach, I will illustrate an example of a compound that inhibits the primary nucleation step in protein aggregation within condensates. These initial results suggest a possible therapeutic route for several neurodegenerative disorders.
