Abstract
In this talk some of the principles thought to govern the sensing of the misfolded/unfolded protein load in the ER and the transcriptional and translational response to that load (referred to collectively as the unfolded protein response or UPR ) will be described. This focus on the physiology of the system will aim to justify the interest in manipulating it to therapeutic ends. Towards the close of the talk, an example of nascent attempts to manipulate the UPR will be discussed.
