Abstract
Plants translate stress stimuli as well as metabolic changes through specific changes (โsignaturesโ) in cytosolic free Ca2+ ([Ca2+]cyt). The channels involved remain poorly characterized. Annexins (Ca2+-dependent membrane-binding proteins) have recently been shown to act as Ca2+-permeable channels in plants. Annexin 1 (ATANN1) of the model plant Arabidopsis thaliana was also implicated in mediating stress signalling in roots and could be acting as a channeI. It is expressed in root epidermal cells and these are accessible to [Ca2+]cyt measurements. Its presence in leaves and its role in drought stress has also been previously shown. We have used yellow cameleon 3.6 (YC3.6) to monitor in vivo differences in [Ca2+]cyt dynamics between wild type and the atann1 knock-out mutant on guard cells and will show preliminary root imaging work.
