Integration of functional genomics and pathway information to elucidate deregulation of signal transduction and drugs' mode of action

Abstract
In this review we will describe recent efforts to integrate signal transduction pathways with various sorts of genomics data, with the aim to find key regulators of gene expression and mechanisms of signal transduction. We will focus on applications related to human disease, and explore how these methods can be used to study drug’s mode of action. Genomic (in particular gene expression) data has been proposed as tool for drug discovery. Here, we will explore how approaches that include prior knowledge on the underlying biochemical processes provide deeper mechanistic insight and enhance the power of genomic data to develop novel therapies for diseases such as cancer.
