3-D cryo-EM in biology: specimen preparation and imaging for tomography

Abstract
Electron tomography of frozen-hydrated specimens, without chemical fixation, staining, or dehydration, allows us to study the 3-D structure of cells and tissue in a near-native state. Preparation and 3-D imaging of such specimens is challenging. Developments in the application of cryo-ultramicrotomy and cryo-FIB, as well as TEM imaging using zero-loss energy filtration and Zernike phase contrast, will be discussed.
