TPP riboswitches: to Chlamydomonas and beyond

Abstract
Thiamine is required for the synthesis of thiamine pyrophosphate (TPP), an essential cofactor for many central respiratory pathways. In most organisms that synthesise thiamine, the level of intracellular thiamine is highly regulated at the level of gene expression, mostly by TPP riboswitches. They are sequences in the mRNA to which TPP binds directly, thereby influencing expression of the transcript. Genetic analysis of riboswitches is well studied in prokaryotes, but limited in eukaryotes. The aim of my PhD project is to characterize TPP riboswitches in Chlamydmonas reinhardtii at a detailed molecular level in order to apply as a powerful tool for synthetic biology.
