Abstract
A number of recent studies using high-throughput single cell gene expression profiling have uncovered a surprising degree of cell-to-cell variability within apparently functionally homogeneous embryonic or induced pluripotent stem cell populations. Collectively, these reports suggest that both deterministic and stochastic mechanisms play key parts in pluripotent stem cell identity. In this talk I will discuss some of the origins and consequences of this cell-cell variability, particularly focussing on the role that feedback loops in the underlying transcriptional regulatory circuitry have in controlling population variability. I will also outline a statistical view in which appropriately defined heterogeneity is a central feature of pluripotent stem cell populations.
