Molecular regulation of skeletal muscle stem cells in distinct cell states

Abstract
The regulatory cell state of founder stem cells during development, and those in the adult is determined by critical transcription factors. We and others have demonstrated that distinct gene regulatory networks operate in different anatomical locations to give rise to skeletal muscle progenitor cells. Despite this diversity, the core cell fate determinants, Myf5, Myod and Mrf4 regulate cell identity in all locations. Upstream of this progenitor population, Notch signaling maintains the stem cell population whilst allowing temporal cell identities to emerge. Cell heterogeneity in the adult muscle stem cell population is observed among quiescent and proliferating cells. We have isolated and characterised subpopulations of muscle stem cells during homeostasis and regeneration. Single cell analysis provides insights into the pallette of behaviours of muscle stem cells.
