Abstract
In vitro, 3D cultures are limited by oxygen and nutrient diffusion rates resulting in necrosis at the centre of large scaffolds. Mass transport in these cultures can be controlled to a large extent by use of dynamic bioreactors. The bioreactor configuration and operation is important in the success of the culture. Previous work has showed biaxial rotation was more successful for in vitro tissue generation than single axis but did not describe the mechanism underlying this effect.
In this presentation, a description of the current state of the bioreactor field for bone growth is described before the key design requirements of a new low cost design are identified. Finally to investigate the parameters that are critical when designing a rotating bioreactor, two low-cost, adaptable and open source bioreactor systems are presented and control systems verified for rotational and positional control.
